Bradford Assay (Quick and Dirty From Kaplan Lab)

1. Dilute Bradford Assay solution with dI H2O 1:4 (in a 15 ml or 50 ml conical tube, for example, and then store @ RT).

2. Put 1 ml of this diluted solution into 1.5 ml microfuge tubes. Prepare tubes in triplicate for each sample to be quantitated.

3. Add 0.5, 1, and 2 ul of sample to each of the three microfuge tubes.

4. Invert to mix.

5. Take spectrophotometric reading at O.D. 595

6. If within the range of 0.1 - 0.8, then do the following calculation:

For 1 ul sample:

O.D. 595 x 40 = _____ mg/ml

For 0.5 ul sample:

O.D. 595 x 40 x 2 = _____ mg/ml

For 2 ul sample:

O.D.595 x 40/2 = ______ mg/ml


If not within the range of 0.1 - 0.8, then adjust amount of sample used and take another reading.
