1. Centrifuge tube containing lyophilized primer.
2. Using a filter-tipped pipet tip, add 100 ul of sterile-filtered water.
3. Rehydrate for 2 minutes.
4. Vortex for 15 seconds.
5. Spin down briefly in "toy" (6-place) microcentrifuge.
6. Transfer primer solution to a 1.5 ml microfuge tube (that has been tagged with a label from the company that coincides with the label that was on the original vial of lyophilized primer).
7. In a separate 1.5ml micfofuge tube, dilute 1:500 with sterile water.
8. Spectrophotometric reading (using 200 ul out of the 500 ul dilution in a quartz cuvette and measuring at O.D.260).
9. Determine concentration.
O.D.260 x 500 x 30 = _______ug/ml
